I agree with Shiemi. They did mention that his entire family was killed during the blue night. So! im thinking that he used the artificial life thing to make her, and somehow implanted memories....But at the same time, doesnt that seem weird to you? What if that actually IS his wife...?
Hmm, as for the whole Paranoia about protecting Rin, I think that it has to do with the reason why he wanted power, which is to protect his brother. I could be wrong though. Maybe Yukio just wants to be able to do something for him? Rin is always saving Yukio, but never the other way around. Maybe he wants to make up for it? Perhaps he feels inadequate because of it xD'' Either way. As for the infection...I think it has to do with Satan for sure now. Maybe it occurs everytime Yukio cant save Rin? maybe Satan did something to him....
And yes. GRANDFATHER?!?!?!?!?! WHAT THE HECK IS THAT SUPPOSED TO MEAN?! He'd have to be on their mothers side though.....

Artificial life would be a good guess, since it's already heavily implied that he's the one behind the factory thing in Siberia the forest.
Yukio's obsessive protection of Rin makes sense, since Rin is tentatively always on the brink of execution (It's been ordered and rescinded what, like four times already?) and since Rin is his brother, and he not only doesn't want to have to kill his brother, but also doesn't want him to become a monster that needs killing. Plus, Rin was the one who was really Shiro's son, though I think Yukio would have rather been the son than the apprentice, so taking care of Rin might help him stay close to Shiro, since he didn't have time to say goodbye and wasn't around for the last bit of time in Shiro's life like Rin was. Kid's got a ridiculous sense of duty, too, so taking care of Rin is probably just routine for him.
The other day (episode, whatever) Wasn't Yukio doing his medical test thing and wasn't there a reaction to one sample of his blood? Based on that, we knew from the start that the rash thingy was demonic in nature somehow. It was somehow connected to his protecting Rin, and preventing him from using his flames, so I really think it's some miasma thing from Rin's flames.
At last, we are getting an exciting baby story! Wait... baby stories aren't exciting. They better be going somewhere good with this...
Gramps could be their "Paternal Grandfather" if Shiro has a father lurking somewhere. I mean, he probably wasn't the result of immaculate conception, and he considered the twins his own pretty much, right?
